Methodenlisting

Auswertung (Statistische Methode) For survival and embryo development data, one-way analysis of variance was used to determine differences among means, with a 0.05 level of significance. Survival and development data were transformed by arcsine square root before analysis. Dunnett’s test was used to determine the no-observed-effect concentration.
Organismus Alter/Lebenstadium Embryo
Organismus Herkunft Carlsbad Aquafarm (Carlsbad, CA, USA). Larger adult mussels (7–10 cm) the source were used to obtain gametes for the embryo–larval development toxicity tests.
Endpunktbestimmung Normal survival and combines both survival and normal larval development (as straight-hinged D-shaped prodissoconch stage I) by comparing the number of normally developed larvae to the number of embryos initially added (as determined by a replicate set of initial density vials).
Sauerstoffgehalt 6.4 - 9.3 mg/L
Analytik der Testsubstanz HPLC, laboratory reporting limit 0.1 mg/L
Lösevermittler <= 1mL/L for any test dilution, and acetone controls (1mL/L1)
Testsystem, Multispezies 4 replicates 1-L glass beakers for each test concentration
Konzentrationen, nominal 0 | 0,25 | 0,5 | 1 | 1,5 | 5 | 15 | mg/l
Konzentrationen, real ND | 0,14 | 0,44 | 0,90 | 1,4 | 4,40 | 13,4 | mg/l
Auswertung (95%-CL) EC50 0.75 (0.72–0.79) mg/L
Lichtstärke yellow fluorescent light (Lambda > 500 nm) to minimize photodegradation
Testmedium Filtered (0.45 µm) natural seawater (salinity ~ 32 ppt), collected from the research pier at Scripps Institute of Oceanography (La Jolla, CA, USA)
Kontrolle (Referenzsubstanz) Copper reference toxicant test (EC50 = 6.68 mg/L)
Lösungsmittelkontrolle, Ergebnis Control normal survival was within test acceptability requirements (>70%) in both solvent and clean seawater negative controls with normal survival of 84.0+/-6.07 and 78.9+/-2.19%, respectively.
